Artocarpus heterophyllus or jackfruit is the national fruit of Bangladesh. It is the most widespread source of the genus. The objective of this study is to characterize the different compounds that were extracted and separated from seeds of Artocarpus heterophyllus and were carried out using different methods using High-Performance liquid chromatography (sometimes referred to as high-pressure liquid chromatography), HPLC, Thin Layer Chromatography ( TLC) to identify an anti-diabetic drug( Vildagliptin ) in the methanolic extract of Artocarpus heterophyllus seeds. Standard in vitro procedures were used to screen possible phytochemical, antibacterial,cytotoxic and anti oxidant activities. Thin layer chromatography, ulra-violet spectroscopy and HPLC were used to detect the presence of various types of compound in methanolic extract and also for identification of a standard drug( Vildagliptin ).Antioxidant effects of sseds were measured by DPPH scavenging assay.Antioxidant effects of mathanolic extract were measured by DPPH scavenging assay. Disc diffusion assay of the methanolic extract was performed to show the antibacterial effect using gram positive,gram negative strains of bacteria and fungi.Then experiment continued again with Thin layer chromatographic analysis of these fractions against two anti-diabetic drugs Vildagliptin and Meropenem which were used as a standard. Thin layer chromatographic analysis of Chloroform fraction indicated to the presence of bioactive compound in this fraction wherein Vildagliptin was a standard. Further investigation is needed to ensure the anti diabetic activity of this chloform fraction.[1] Also cytotxic activity of all seven fractions were done in brian shimp lethality assay. The brine shrimp lethality assay is considered a useful tool for preliminary assessment of toxicity. It has also been suggested for screening pharmacological activities in plant extracts. Different concentration of all the fractions were used in the experiment and the ethyl acetate fraction is found to be severe cytotxic than other fractions, The butanol fraction is least cytotxic among them.The methanolic extract showed stronger antioxidant,antidiabetic and slight antibacterial activity. Further investigation is needed for the proper identification and isolation of these bioactive compounds to produce safer drugs for treatment of harmful diseases. |
